近日,某院眼科组实验人员采购高强度紫外线手电筒XEPU-1530用于建立UVA紫外线诱导的体外FECD模型,并在实验中取得理想的效果。
角膜内皮是一层排列有序的角膜内皮细胞(CECs),在维持角膜透明性方面发挥着不可或缺的作用。富克斯角膜内皮营养不良 (Fuchs endothelial corneal dystrophy, FECD)是内源性角膜内皮营养不良的主要病因,其特征为细胞形态改变、细胞数量进行性减少以及细胞外基质赘生物(角膜后弹力层膨出物)形成,最终导致角膜盲。
紫外线(ultraviolet UV)、吸烟、营养和激素影响等环境因素是FECD发病机制中的关键因素。尤其是紫外线,可能会增加细胞的病理应激,影响FECD的发生和发展。目前FECD尚无药物治疗方法,所以建立紫外线诱导的FECD模型并对FECD发病机制进行研究是十分重要的课题。高强度聚光紫外线手电筒XEPU-1530光斑聚集、简单好用,是体外FECD建模的理想紫外光源。
建立UVA紫外线诱导的体外FECD模型的大致流程如下:
将小鼠随机分成5组,每组4-5只小鼠。UV组小鼠角膜接受紫外线照射,对照组不接受紫外线照射。高强度紫外线手电筒XEPU-1530,可发射超高强度的365nm长波紫外线,形成一个聚集的紫外线光斑,非常适合照射小鼠角膜,建立体外FECD模型。

The corneal endothelium is a layer of orderly arranged corneal endothelial cells (CECs) that plays an indispensable role in maintaining corneal transparency. Fuchs endothelial corneal dystrophy (FECD) is the main cause of endogenous corneal endothelial dystrophy, characterized by cell morphological changes, progressive reduction in cell number, and formation of extracellular matrix neoplastic lesions (protrusions of the posterior elastic layer of the cornea), ultimately leading to corneal blindness.
Environmental factors such as ultraviolet (UV) radiation, smoking, nutrition, and hormonal influences are key factors in the pathogenesis of Fundus Entropion and Ectropion (FECD). UV radiation, in particular, may increase cellular pathological stress and affect the occurrence and progression of FECD. Currently, there is no pharmacological treatment for FECD. Therefore, the establishment of UV-induced FECD models and the investigation of its pathogenesis are very important.
